Mechanism of action and preclinical data

• Privosegtor is a peptoid small molecule that crosses the blood-brain and retinal barriers, promoting neuro-axonal survival in multiple in vitro and in vivo models, including glaucoma, MS, and optic neuritis.

• Demonstrated neuroprotection in preclinical models, showing preservation of retinal ganglion cells, reduced optic nerve demyelination, and decreased axonal loss.

• Activates SGK pathways, inducing protective microglia phenotypes and promoting oligodendrocyte differentiation and myelination.

Clinical trial design and patient characteristics

• Phase 2 ACUITY trial was a randomized, double-masked, placebo-controlled, multi-center study in patients with unilateral optic neuritis of demyelinating origin.

• 36 patients were randomized; key endpoints included safety, low contrast visual acuity (LCVA), and retinal structure (GCIPL and RNFL thickness).

• Baseline characteristics were balanced between groups, with similar age, gender distribution, and disease-modifying therapy use.
